Learning outcomes

The course will provide basic knowledge about mouse functional genomics
and complex genetic phenomena that do not follow standard mendelian
inheritance

Course contents

MOUSE STRUCTURAL AND FUNCTIONAL GENOMICS.

Lectures 1-6. The mouse (Mus musculus) genome project.
-A physical map of the mouse genome. Cloning and sequencing strategies. Methods:
cell hybrids, radiation cell hybrids, cloning vectors (Yac, Bac), shot-gun
cloning and sequencing; STS; contigs; automated sequencing
-A transcriptional map of the mouse genome. Methods: Northern blot;
exon-trapping, RT-PCR, CpG island prediction and analysis;
bioinformatic prediction of gene functions.
-The mouse transcriptome. Methods: construction of cDNA library and annotation of ESTs
-Genome Anatomy: Global transcription profiles in mouse development and
tissue differentiation. Methods: cDNA microarray; Serial Analysis of
Gene Expression; RNA-RNA Tissue in situ hybridization

Lectures 7-12. Gene targeting in mouse.
-Knockout mice; conditional knockout mice, knockout through gene trapping.
-Transgenic mice, conditional transgenic mice
-Chromosome engineering in mice. Constructing balancer chromosomes in mice. Genome-wide ENU mutagenesis screen in mice

Lecture 13-20 Epigenetic mechanisms and complex phenotypes.
-X chromosome inactivation
-Genomic imprinting
-Small RNAs (microRNAs and siRNAs) as gene expression modulators.
-Epigenetics in cancer and in specific monogenic inherited disorders (ICF, Rett, FSHD, Rubinstein-taybi)
